Team Tu Delft Safety 2013 Igem Org Safety the mrsa (methicillin resistant staphylococcus aureus) problem is our inspiration for this project. this project will focus on killing the staphylococcus species by antimicrobial peptides synthesized in e. coli. as we work in the lab which has only bsl 1 permit we did intense literature study to find closely related organisms of same genera other than s. aureus (dsmz 6148) that can be. We are the igem 2013 team of delft university of technology, netherlands. we are working on the production of antimicrobial peptides in order to kill methicillin resistant staphylococcus aureus, mrsa.

Team Tu Delft 2013 Igem Org In order to ensure the safety of the system, the peptide activating protease is combined with a kill switch that will kill our e. coli (figure 1). this project is novel because we aim to fight a life threatening, hard to cure disease by incorporating and combining components of multiple kingdoms into one organism. By using the band aid, the safety of the real application can be ensured in several ways. first of all, the complete system is enclosed by the band aid and in that way it can be located on the wound without having the e. coli in direct contact with the patient which could lead to the e. coli also becoming a source of infection. General lab safety we are very lucky to have had our very own lab space during igem; a cozy ml 1 lab in the new applied sciences building of the tu delft campus. ml 1 is the lowest safety level when working with genetically engineered microorganisms, meaning that experiments involve low or no risks (e.g. no human pathogens). Biosafety regulations all the members of our igem team are students of the technical university of delft. throughout the months that we are working on our project, we of course work a lot on the lab. our lab is part of the tu delft faculty of applied sciences and, in particular, of the kluyver laboratory for biotechnology.

Kill switch to the circuit a kill switch is added for two reasons: bio safety and secretion. the bio safety concern is that the e.coli is in essence a treat to the humans and thus it is killed after it has done what it is supposed to. the second reason is the efficient secretion: if the peptides are not secreted naturally it is very difficult to force them. the solution is cell lysis, then all.
